RESUMO

OBJECTIVE To establish methods to identify the chemical components of Gantaishu capsule, and determine the contents of 6 index components including glycyrrhizic acid. METHODS The chemical components of Gantaishu capsule were determined by HPLC-TOF/MS; the contents of 6 index components including glycyrrhizic acid were determined by UPLC-MS/MS. RESULTS A total of 41 chemical components were identified in Gantaishu capsules. The linear ranges of glycyrrhizic acid, mangiferin, luteolin, costunolide, oleanolic acid and berberine were 200-10 000 ng/mL(r were all greater than 0.999). The limits of quantification were 200, 20, 10, 1, 10, 0.5 ng/mL, and the limits of detection were 100, 10, 5, 0.5, 5, 0.25 ng/mL, respectively; RSDs of precision, stability (24 h) and reproducibility tests were all less than 5.0% (n=6 or n=3); the recoveries were 99.05%-101.08% (RSD were all less than 2.0%, n=6). The contents of them were 2.42-2.66, 0.85-1.16, 0.35-0.46, 6.18- 6.46, 0.99-1.29, 5.22-5.56 mg/g. CONCLUSIONS The established methods for identification and content determination are rapid and simple, and can be used for the identification of chemical components and the content determination of index components in Gantaishu capsule.

RESUMO

Objective To study the correlation of the adiponectin receptor 1 gene rs1139646C/G and rs10920531C/A single nucleotide polymorphisms with Chinese Han women with polycystic ovary syndrome (PCOS).Methods For PCOS group (n =80) and control group (n =80),polymerase chain reaction (PCR) was used to amplify the upstream and downstream 250 bases of rsl139646C/G and rs10920531C/A,respectively.PCR products were directly sequenced.Results For rs1139646C/G loci sites between PCOS and control groups,all were the homozygous genotype CC,and no statistical significance was found for genotype frequency and allele frequency between two groups.For rs10920531C/A loci sites between PCOS and control groups,no statistical significance was found for genotype frequency and allele frequency between two groups (P =0.683,0.580),and no statistical significance was found for two loci with genetic linkage disequilibrium analysis (x2 =0.307,Fisher 's P =0.579610,Pearson' s P =0.579608).Conclusions Adiponectin receptor 1 gene rs1139646C/G and rs10920531C/A single nucleotide polymorphisms have nothing to do with Chinese Han PCOS women.Genetic linkage disequilibrium analysis has nothing to do with PCOS.The rs1139646C/G in Chinese Han population is relatively stable site,and no locus mutation is found.

RESUMO

Objective To study the influence of local administration of Icariin on orthodontically induced inflammatory root resorption (OIIRR ).Methods After rat models of orthodontic root movement were successfully established,rats were injected with 200 mg/kg Icariin (Icariin group)or saline (positive control group), respectively.Left maxillary served as the negative control group.The resorption pit of root surface was measured using scanning electron microscopy (SEM), HE staining, and tartrate resistant acid phosphatase (TRAP ). Results The level of TRAP positive cells and root resorption score in Icariin group were significantly lower than those in the positive control group (P 0.05).Conclusion Local administration of Icariin may reduce the degree of orthodontically induced resorption.

RESUMO

Objective:To survey the relation of different vertical skeletal facial type and the third molar agenesis.Methods:The vertical face type of 207 subjects was divided into high angle(n=45),average angle(n=99)and low angle(n=63)groups by the assessments of MP-SN angle.The assessment of the third molar was made from panoramic radiographs,then the third molar agenesis rate of different vertical skeletal facial types was analysed.Chi-square criterion examination was conducted with SPSS 17.0 software. Results:In the high angle group,whether in the maxillary or mandibular,unilateral or bilateral,the absent percentage of third molar was higher than that in low angle group(P<0.05).In the females the bilateral maxillary third molar absent percentage was higher than in the males(P<0.05).The absent percentage of maxillary third molar was higher than that of the mandibular third molar(P<0.05).Conclusion:The third molar absence depends on vertical skeletal facial type.The absent percentage of third molar in the in-dividuals with high angle type is higher than that in those with low angle type(P<0.05).

BACKGROUND: Icari n as one of the main components of Epimedium has an inhibitory effect on osteoclasts.. OBJECTIVE: To further investigate the influence of icariin on the root absorption of the maxil ary first molar at mesial part during orthodontic treatment in rats. METHODS: Orthodontic root resorption models were established in the left maxil a of rats. Local injection of 200 mg/kg icari n (icari n group) or normal saline (positive control group) was administrated into the left first molar buccal periosteum. The right maxil a of rats served as negative control group that was treated with neither appliance nor drug injection. The mesial distance between bilateral first molars and the contralateral maxil ary incisor was measured before and after the appliance was placed. Mesial surface of the mesial root of bilateral maxil ary first molars was observed using scanning electron microscope. RESULTS AND CONCLUSION: Mesial movement of the maxil ary molars in the icari n group was significantly less than that in the positive control group (P < 0.05). Under the scanning electron microscope, smal absorption lacunae were scattered in the icari n group, while the positive control group showed a large amount of absorption lacunae and they were interconnected into a sheet, showing a stark contrast with the smooth root surface of the negative control group. It is indicated that icari n can inhibit root resorption caused by orthodontic treatment, while reducing the amount of mesial movement of the molar under corrective force.

RESUMO

Objective To explore the effect of domoxifen combined with tamoxifen on ovulation in clomiphene-resistant patients with polycystic ovary syndrome. Methods 100 patients who was resistant to CG were randomly divided into two groups:the treatment group (group A) and control group (group B). Oral dosing CC 100mg/d to the patients of group A in the first 3th to 7th days of menses,at the same time,oral dosing tamoxifen 40mg/d in the first 3th to 7th days of menstrual cycle. The dose of CC to group B was same as group A. Beginning to monitor the case of follicular growth during the 8th day of menses. When a ovarian follicle' s diameter beyond 18mm or 2 ovarian follicle' s diameter beyond 17mm, intramuscular injection hCG 10 000IU, sexing after 36 hours later. Observing the two groups of the mature ovarian number,the endometrial thickness, the days of promoting ovulation ,the level of progesterone in serum about the 22th day of menstrual cycle, ovulation rate and pregnancy rate, after injecting hCG.Results The day after injecting hCG, the mature ovarian follicle number,the endometrial thickness, the level of progesterone in serum about the 22th day of menstrual cycle,ovulation rate and pregnancy rate,group A was significantly different from group B( P <0.05 ~ P <0. 01 ) ,the days of promoting ovulation have no statistically significant differences between the groups (P > 0. 05). Conclusion In the clomiphene-resistant patients with polycystic ovary syndrome,tamoxifen could promote ovulation,improve the effect of CC on the uterus' s anti-estrogen,increase endometrial thickness, elevate ovulation rate and pregnancy rate.
